Heme biosynthesis pathway Two different ways of 5-Aminolevulinate (ALA) synthesis Remaining 7 steps are conserved among all organisms Photosynthetic organisms use the same pathway for the Chlorophyll synthesis Eukaryotes differ in the origin of the genes and intracellular localisation of the enzymes
Chlorophyll Biosynthesis in Photosynthetic eukaryotes In the Photosynt. Cell, the majority of the end-products are needed in the Plastid Chlorophyll is synthesized in much higher rate than the Heme. Most Heme is used in the Plastid for cytochromes and for synthesis of Bilin Chromophores Only small portion of the heme is needed in mitochondrion for respiratory complexes
Chlorophyll Biosynthesis in Chromera velia Chromera possesses ALAsynthase that is normally found only in heterotrophic eukaryotes Chromera velia is the only alga studied so far that is able to synthesize Chlorophyll a from Glycine and Succinyl-CoA

The role of carotenoids is not only to transfer energy, but also to protect the cell against damage by O 2 at high light intensities. If the antenna is flooded with light too rapidly for the reaction centers to be reoxidized, then the antenna chlorophylls discharge most of the extra energy as fluorescence and heat. However, the excited molecules also have an opportunity to evolve into an excited triplet state, inwhichthe spins of the two unpaired electrons are parallel (Fig. a). These excited triplet states are relatively long-lived. They cannot decay to the ground state (singlet) unless the electronic spin changes again, and this does not happened readily. reaction with O 2 The phosphorescence is too slow compared to a reaction between the triplet 3 Chl* and O 2
One way that they do decay is by reacting with O 2 molecules which has a triplet ground state (see chap 14, slide 18). The reaction returns the chlorophyll to its ground state and promotes the O 2 to an excited singlet state (Fig. b) This change can have lethal consequences for the cell because singlet O 2 is extremely toxic. It reacts irreversibly with a variety of groups in proteins, nucleic acids and lipids. Carotenoids (Car) prevent these destructive side reactions by quenching the excited triplet chlorophyll before it has a chance to react with O 2 (Fig. c). In this process, the chlorophyll returns immediately to its ground state, and the carotenoid is elevated to an excited triplet state. The carotenoid triplet state cannot generate singlet O 2 because it lies below the singlet O 2 in energy. Instead, it decays harmlessly to the ground state. Carotenoids also can quench singlet O 2 itself.
